Purpose of use

NKCult Kit (Catalog Number: 298) is specifically designed for the isolation, activation, and expansion of Natural Killer (NK) cells from a small volume of peripheral blood (as low as 20 mL).

The product is intended for direct application in the field of Advanced Therapy Medicinal Products (ATMPs). It provides a comprehensive solution for GLP/GMP-compliant laboratories engaged in the manufacturing of safe “cell-based medicinal products,” meeting stringent clinical release criteria, including: cell viability > 90%, phenotypic purity > 80% (CD3⁻CD56⁺ population), and potent cytotoxic activity against cancer cells (> 50%).

Description

Based on the core principle of regenerative medicine - “Cells are the Medicine” - the NKCult Kit is not merely a reagent kit, but an integrated, all-in-one technological ecosystem designed with strict adherence to the physiological characteristics of Natural Killer (NK) cells:

Maximum safety with a serum-free platform:
The nutrient source is completely free of animal-derived serum, thereby eliminating risks of contamination and xenogeneic immune reactions.

Spatially oriented 3D activation:
The process is initiated using a surface-coated recombinant protein system (Coating Reagent), providing specific recognition signals that promote cell adhesion and the formation of natural proliferative clusters without the use of feeder cells.

Phased nutrition strategy:
A specialized micronutrient system is tailored for the receptor priming phase (Day 0–3) and the rapid expansion phase (Day 4–21). Associated consumables are optimized to protect cells from mechanical (shear) stress and thermal shock.

Flexible harvesting strategy:
The kit enables clinical decision-making through two optimized harvesting scenarios:

  • “Peak potency” scenario (Day 16): Harvest NK cells with enhanced tumor-homing capacity and superior in vivo persistence.
  • “Maximum yield” scenario (Day 21): Intended for protocols requiring ultra-high infusion doses, enabling expansion to billions of cells.

Components

Product Code 298 is precisely calibrated and standardized for a single culture batch derived from 20 mL of peripheral blood, including:

Group I: Reagents and Biological Media

  • NKCult Basal (298-1): 1 bottle (540 mL) – Serum-free basal medium providing essential nutrients.
  • NKCult Coating Reagent (298-2): 1 vial (0.5 mL) – Recombinant protein used for activation surface coating.
  • NKCult D0–3 Supplement (298-3): 1 vial (2.0 mL) – Micronutrient system for early-stage receptor activation.
  • NKCult D4–21 Supplement (298-4): 5 vials (8.0 mL/vial) – Micronutrient system supporting proliferation and large-scale expansion.

Group II: Specialized Consumables

  • MNC Collection Tube (298-7): 3 tubes (8 mL/tube) – Pre-filled with ACD-A anticoagulant, optimized for mononuclear cell isolation.
  • T-75 Flask, Non-treated (298-5): 1 flask – Specialized surface for protein coating and activation.
  • NKCult Bag 500 mL (298-6): 1 bag – Gas-permeable culture bag designed for scale-up expansion.

Storage Conditions

This product contains highly sensitive recombinant protein growth factors and MUST be stored under strictly controlled temperature conditions:

  • Frozen storage (≤ -20°C): Applicable to high-activity components (NKCult Coating Reagent, NKCult D0–3 Supplement, NKCult D4–21 Supplement).
  • Refrigerated storage (2°C to 8°C): Applicable to NKCult Basal medium.
  • Controlled room temperature (dry, well-ventilated): Applicable to plastic consumables (T-75 flask, NKCult bag, MNC tubes).

[⚠️ Critical Control Point – CCP]:
The fully prepared D4–21 expansion medium (Basal + Supplement) MUST be stored at 2°C to 8°C and maintains optimal activity for 7 to 14 days only. In accordance with ATMP principles: prepare only the quantity required for immediate use.


Shipping Conditions

To preserve the structural integrity of recombinant proteins, the kit MUST be transported in strict compliance with cold chain logistics:

  • Frozen components (≤ -20°C): Must be shipped on dry ice within validated insulated containers, maintaining a frozen state throughout transit.
  • Refrigerated components (2°C to 8°C): Shipped with ice packs/gel packs, with appropriate thermal barriers to prevent direct contact and avoid thermal shock or localized freezing.
  • Consumables: Shipped at ambient temperature with protective, impact-resistant packaging. Particular care must be taken to avoid deformation of the gas-permeable membrane of the culture bag.

Mechanical handling precautions:
Avoid excessive agitation, dropping, or centrifugal forces that may disrupt the delicate structural integrity of recombinant protein components within the vials.
